KVA12402 is a fully human monoclonal antibody with an IgG4 backbone that targets V-domain Immunoglobulin Suppressor of T cell Activation (VISTA), also known as PD-1H. Developed by Kineta Inc., KVA12402 functions as an immune checkpoint inhibitor. VISTA is a negative checkpoint protein highly expressed on myeloid cells and leukemic blasts, where it acts to suppress anti-tumor T cell responses. Preclinical data presented at ASH 2023 demonstrated that KVA12402, both as a monotherapy and in combination with standard chemotherapy (cytarabine and doxorubicin), significantly reduced tumor burden and improved survival in disseminated models of Acute Myeloid Leukemia (AML). It is a variant of Kineta's lead VISTA-targeting program, KVA12123, which utilizes an IgG1 backbone.
