Overview

KVX-053 is a novel, potent, reversible small molecule inhibitor of protein tyrosine phosphatase 4A3 (PTP4A3/PRL-3), developed as an anti-inflammatory and antineoplastic agent. It acts as a selective allosteric inhibitor of PTP4A3 and also inhibits PTP4A2. Preclinical studies have demonstrated that KVX-053 has significant immunomodulatory and anti-inflammatory effects by blocking the activation of multiple inflammatory pathways. In animal models of acute lung injury (ALI) and acute respiratory distress syndrome (ARDS), including those induced by SARS-CoV‑2 spike protein or chemical agents like CEES (a sulfur mustard analog) or LPS (lipopolysaccharide), KVX‑053 reduced pulmonary inflammation, vascular leakage, cytokine overexpression, structural lung injury and improved lung function. The drug is being developed primarily for conditions involving pulmonary microvascular leakage and inflammation such as ALI/ARDS but has also been investigated in preclinical models for cancer indications including ovarian cancer and glioblastoma[1][2][6].
