Overview

KW-120-94-1 is a synthetic analog of the natural product withaferin A, which is derived from the plant *Withania somnifera* (Ashwagandha). Developed and studied at the University of Arizona, this compound has demonstrated potent anti-cancer activity in preclinical models of pancreatic cancer, including poorly-differentiated (Panc-1), moderately differentiated (MiaPaca-2), and well-differentiated (BxPC-3) cell lines. Its mechanism of action involves the up-regulation of the mitogen-activated protein kinase (MAPK) signaling pathway and the glutathione metabolic pathway, leading to increased cellular stress and the induction of apoptosis. KW-120-94-1 has shown growth inhibitory effects with IC50 values ranging from 0.45 to 2.25 µM across different pancreatic cancer cell lines, characterized by morphological changes and PARP cleavage.
