KW‑2450 is an orally bioavailable small molecule that acts as a dual inhibitor of the insulin-like growth factor 1 receptor (IGF‑1R) and insulin receptor (IR) tyrosine kinases. By selectively binding to and inhibiting these receptors, KW‑2450 blocks downstream signaling pathways involved in tumor cell proliferation and survival, leading to inhibition of tumor growth and induction of apoptosis. Preclinical studies demonstrated its activity against various cancer cell lines, particularly in combination with other agents such as lapatinib or letrozole. Clinical development has focused on advanced/metastatic hormone receptor-positive, HER2-positive breast cancer, but trials have not progressed beyond early phases[1][3][4][5][6].
