KY102 is a novel nanobody-drug conjugate (NDC) targeting Glypican-3 (GPC3), a cell-surface oncofetal glycoprotein highly expressed in hepatocellular carcinoma (HCC). Developed by Hangzhou Kunyu Pharmaceutical, KY102 consists of a monovalent nanobody covalently conjugated to a topoisomerase 1 inhibitor payload, DXD (deruxtecan), via a cleavable glycyl glycyl phenylalanyl glycine (GGFG)-aminomethyl (AM) linker. It is designed with a drug-to-nanobody ratio (DAR) of approximately 3. Preclinical studies demonstrate that KY102 has high binding affinity to GPC3 across multiple species (human, cynomolgus monkey, rat, and mouse) and undergoes rapid internalization in GPC3-expressing tumor cells. It has shown potent, target-specific cytotoxicity and effective bystander-mediated killing in HCC models. In vivo, KY102 demonstrated robust tumor growth inhibition in cell line-derived and patient-derived xenograft models with encouraging tolerability. An Investigational New Drug (IND) application is anticipated in 2026.
