KY19334 is a first-in-class small-molecule inhibitor developed by CK Regeon that targets the protein-protein interaction (PPI) between CXXC-type zinc finger protein 5 (CXXC5) and Dishevelled (Dvl). CXXC5 acts as a negative feedback regulator of the Wnt/beta-catenin signaling pathway by binding to the PDZ domain of Dvl, thereby suppressing pathway activation. By blocking this interaction, KY19334 restores suppressed Wnt/beta-catenin signaling, which is a critical pathway for tissue regeneration and metabolic homeostasis. The drug is being developed for a wide range of indications where Wnt signaling is impaired, including androgenetic alopecia (hair loss), diabetic wound healing, obesity, type 2 diabetes, and neurodegenerative conditions such as Alzheimer's disease. Preclinical studies have demonstrated its potential to promote hair regrowth, accelerate wound closure, and improve metabolic parameters.
