Overview

KYP-2047 is a potent, selective, and blood-brain barrier-permeable small-molecule inhibitor of **prolyl oligopeptidase (PREP)**, also known as prolyl endopeptidase. It is frequently utilized as a reference compound in preclinical research focusing on neurodegenerative disorders, particularly **Parkinson's disease** and **Alzheimer's disease**. The mechanism of action involves the inhibition of PREP, an enzyme that has been shown to accelerate the aggregation of **alpha-synuclein**, a protein central to the pathology of Parkinson's. By inhibiting PREP, KYP-2047 can reduce the formation of toxic alpha-synuclein oligomers and enhance autophagic clearance. While it has demonstrated efficacy in reducing protein aggregation in various models, recent comparative studies suggest it may have limited effects on neuroinflammation and behavioral recovery compared to next-generation PREP ligands.
