Overview

KYV-201 is an investigational allogeneic anti-CD19 chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of B-cell driven autoimmune diseases. It utilizes a fully human anti-CD19 CAR construct, identical to that used in the autologous product KYV-101, but is manufactured using a differentiated allogeneic platform developed by Intellia Therapeutics. This platform incorporates CRISPR/Cas9-mediated gene edits to remove the endogenous T-cell receptor and key cell surface proteins, aiming to prevent graft-versus-host disease and enhance immune evasion for prolonged CAR T-cell persistence. The therapy targets CD19-expressing B cells, leading to their depletion and potentially resetting immune function in conditions such as lupus nephritis, multiple sclerosis, myasthenia gravis, systemic sclerosis, and other B-cell mediated autoimmune disorders[1][2][3][5].
