KZR-261 is a first-in-class, small molecule anti-tumor agent developed by Kezar Life Sciences. It acts as a direct inhibitor of the Sec61 translocon, a protein complex involved in the translocation of newly synthesized proteins into the endoplasmic reticulum. By inhibiting Sec61 activity, KZR-261 disrupts protein secretion pathways that are essential for tumor cell survival and proliferation. The drug was designed to have broad-spectrum anti-tumor activity and was evaluated in Phase 1 clinical trials for advanced solid malignancies including melanoma (and uveal melanoma), mesothelioma, colorectal cancer, prostate cancer (castrate-resistant), and other metastatic or unresectable solid tumors[4][5][6][8]. Development has been discontinued following completion of Phase 1 studies[4].
