Overview

KZR-8834 is a novel, small molecule inhibitor of the Sec61 translocon, the primary channel for protein translocation into the endoplasmic reticulum (ER). Developed by Kezar Life Sciences, it selectively blocks the secretion and membrane insertion of specific "client" proteins, including growth factors and oncogenic transmembrane proteins, while maintaining a favorable safety profile compared to earlier Sec61 inhibitors. In preclinical studies, KZR-8834 demonstrated potent anti-tumor activity across various malignancies, including multiple myeloma, melanoma, and head and neck squamous cell carcinoma, by inducing rapid apoptosis through caspase activation. It has shown efficacy in mouse xenograft models both as a monotherapy and in combination with other agents, suggesting a broad therapeutic index for treating solid and hematologic cancers.
