L-ICON3

L-ICON3 is a third-generation tissue factor–targeting immune conjugate platform being developed by Eikonoklastes Therapeutics as an immunotherapy for solid tumors, initially focused on triple-negative breast cancer and other high-unmet-need cancers.[4][11][15][17] Engineered by Zhiwei Hu as an evolution of earlier ICON and L-ICON1 (L-ICON) constructs, L-ICON3 is designed to bind tissue factor expressed on tumor cells, tumor neovasculature, and tumor stem cells, thereby directing immune effector mechanisms to simultaneously attack multiple components of the tumor microenvironment and stimulate the body’s natural antitumor immune response.[7][11][13][15] The platform seeks to improve efficacy and safety over prior generations by optimizing targeting, effector engagement, and pharmacologic properties, and is currently in preclinical/IND-enabling development with no approved indications.[4][11][15][17]
