Overview

l-pipecolic acid as-APF is a synthetic derivative of antiproliferative factor (APF), which itself is a frizzled protein 8-related sialoglycopeptide known to inhibit epithelial cell proliferation, particularly in bladder carcinoma and other tumor types. The l-pipecolic acid *as*-APF variant incorporates l-pipecolic acid into the peptide backbone of the APF derivative. It demonstrates potent, dose-dependent inhibition of proliferation across a wide range of carcinoma and melanoma cell lines, apparently independent of cell type or mutation status (p53, ras). Mechanistically, APF and its analogs, including l-pipecolic acid *as*-APF, act by binding to cytoskeletal associated protein 4 at the cell membrane, resulting in inhibition of cell proliferation, suppression of HB-EGF and matrix metalloproteinase 2 (MMP2) production, and attenuation of in vitro cell invasion[1].
