l-tetrahydrofuroic acid (L-THFA) is a competitive inhibitor of proline dehydrogenase (PRODH, also known as proline oxidase), a mitochondrial inner membrane flavoprotein that catalyzes the first step of proline catabolism. By mimicking the structure of L-proline, l-tetrahydrofuroic acid reversibly binds to the active site of PRODH, blocking the FAD-dependent oxidation of proline to pyrroline-5-carboxylate (P5C). This inhibition disrupts proline-derived mitochondrial ATP production and reactive oxygen species (ROS) generation. Preclinical studies have investigated l-tetrahydrofuroic acid as a chemical probe to evaluate the role of proline catabolism in cancer cell survival, metastasis, and fungal morphogenesis, demonstrating its ability to reduce breast cancer metastasis in mouse models.
