Overview

L-Vax + Bacillus Calmette-Guérin + cyclophosphamide is a combination regimen designed as an investigational cancer immunotherapy. L-Vax refers to an autologous tumor cell vaccine; Bacillus Calmette-Guérin (BCG) is a bacterial immunostimulant often used as an adjuvant to activate immune responses against tumors; cyclophosphamide is a cytotoxic alkylating agent with both antineoplastic and immunosuppressive activities. Cyclophosphamide works by alkylating DNA, causing cross-links and inhibition of tumor cell replication, and modulates the immune system by selective T-cell depletion and cytokine modulation[2][4][6][8]. BCG acts as a non-specific immune adjuvant that enhances the body’s immune response against tumor antigens present in the vaccine component[3][5]. The combination is intended to boost anti-tumor immunity and enhance chemoimmunotherapy efficacy by combining immune activation (BCG, L-Vax) with cytotoxic cell reduction (cyclophosphamide)[1][5].
