L001 is a novel, potent, and selective small molecule antagonist of the prostaglandin E2 receptor 4 (EP4). It is being investigated for its therapeutic potential in metastatic pancreatic cancer. L001 has demonstrated effectiveness in repressing PGE2-elicited cell migration and invasion of pancreatic cancer cells in a dose-dependent manner. In preclinical models, L001, both alone and in combination with gemcitabine, showed significant anti-metastasis activity in a pancreatic cancer hepatic metastasis model with good tolerability and safety. Its mechanism involves abrogating Yes-associated protein 1 (YAP)-driven pro-metastatic factor expression.
