Overview

L16-TNF is an immunocytokine composed of a human single-chain antibody fragment (scFv), designated L16, fused to human tumor necrosis factor-alpha (TNFα). The L16 moiety specifically targets the L1 cell adhesion molecule (L1CAM, also known as CD171), a glycoprotein frequently overexpressed in various aggressive cancers, including neuroblastoma, melanoma, and ovarian carcinoma, while showing limited expression in normal tissues. By concentrating TNFα at the tumor site, L16-TNF aims to induce localized hemorrhagic necrosis, activate anti-tumor immune responses, and disrupt tumor vasculature, thereby overcoming the systemic toxicity that limits the clinical use of free TNF. Developed by Philogen, this asset has demonstrated potent anti-tumor activity in preclinical models, both as a monotherapy and in combination with other agents.
