Overview

L17-F05 is a novel lipid nanoparticle (LNP) formulation designed for the efficient delivery of mRNA vaccines, particularly in the context of cancer immunotherapy. Identified through a systematic screening of ionizable lipids and LNP compositions using human primary antigen-presenting cells (APCs), L17-F05 exhibits unique self-adjuvanting properties. It functions by activating the stimulator of interferon genes (STING) pathway specifically within lymph node macrophages, which in turn triggers a type-I interferon (IFN-I) response. This innate immune activation promotes the maturation of conventional dendritic cells (cDCs) and enhances the presentation of tumor antigens to T cells. Preclinical studies have demonstrated that L17-F05 mRNA vaccines encoding melanoma-associated antigens, such as Gp100 and Trp2, can effectively inhibit tumor growth and prolong survival in mouse models.
