Overview

L19-2C11 is a bispecific T-cell engaging antibody (BsAb) designed for cancer immunotherapy, specifically targeting the extra domain B (EDB) of fibronectin and the CD3 epsilon subunit of the T-cell receptor. Developed by Philochem and Philogen, the molecule features a unique architecture consisting of a Fab fragment (targeting CD3 via the 2C11 clone) fused to two single-chain Fv (scFv) fragments (targeting EDB via the L19 clone) at the C-terminus. EDB is a marker of tumor angiogenesis and stroma, making it a pan-tumoral target. L19-2C11 is intended to bridge T cells to the tumor microenvironment to induce localized immune activation. Preclinical studies in immunocompetent mouse models have demonstrated that while the antibody effectively localizes to EDB-rich stroma, its therapeutic efficacy is significantly higher when the target is expressed on the tumor cell surface, suggesting that direct cellular targeting may be superior to stromal targeting for T-cell engaging bispecifics.
