L1CAM antibody refers to a class of preclinical therapeutic agents, including monoclonal antibodies and antibody-drug conjugates (ADCs), designed to target the L1 cell adhesion molecule (L1CAM). L1CAM is a transmembrane protein frequently overexpressed on metastasis stem cells (MetSCs) and glioma stem cells, where it facilitates cell motility, epithelial-mesenchymal transition (EMT), and angiogenesis. In the ADC format, these agents typically consist of a monoclonal IgG1 antibody conjugated to a potent cytotoxic payload, such as the anthracycline derivative PNU-159682, via cleavable or non-cleavable linkers. The mechanism of action involves binding to the extracellular domain of L1CAM, leading to receptor-mediated internalization and intracellular release of the payload, or blocking the interaction between L1CAM and integrins (such as αvβ3) to inhibit tumor cell migration and vascular recruitment. Development is primarily occurring in academic and research institutions, such as Weill Cornell Medicine and the Cleveland Clinic, with a focus on aggressive solid tumors including triple-negative breast cancer, lung adenocarcinoma, pancreatic ductal adenocarcinoma, and glioblastoma.
