L2-8R-70 CAR T cells are an experimental "3-in-1" chimeric antigen receptor (CAR) T-cell therapy developed by researchers at the University of Florida to treat aggressive solid tumors, specifically glioblastoma multiforme (GBM). This next-generation construct is engineered with three distinct functional modules: a CAR targeting the CD70 antigen on tumor cells, the expression of the IL-8 receptor (CXCR2) to enhance T-cell trafficking into the tumor microenvironment (TME), and the secretion of soluble LAIR2. LAIR2 acts as a decoy receptor to inhibit Leukocyte-associated Ig-like receptor 1 (LAIR1) signaling, which is typically triggered by collagen in the TME to promote immunosuppressive M2-like tumor-associated macrophages (TAMs). By disrupting the LAIR1/FXIII-A/collagen IV pathway, these CAR T cells aim to remodel the TME, reduce collagen-mediated physical barriers, and enhance the infiltration and effector function of immune cells in "cold" tumors.
