Overview

L218CAR19 is a chimeric antigen receptor T-cell (CAR-T) immunotherapy that specifically targets CD19 on B cells[1][2][3]. This therapy is being developed by Henan Cancer Hospital and is currently in Phase 1 clinical trials for patients with relapsed or refractory B-cell lymphomas[1][2][3]. ## Mechanism of Action L218CAR19 functions as a CD19 modulator, specifically targeting the B-lymphocyte antigen CD19 on malignant B cells[2]. As a CAR-T cell therapy, it works by genetically modifying a patient's own T cells to express a receptor that recognizes CD19, enabling these modified T cells to target and destroy cancer cells that express this antigen. ## Clinical Development The therapy is currently being evaluated in a Phase 1, open-label, dose-escalating study (NCT06478381) that began recruiting in June 2024[1][3][8]. This trial aims to assess the safety, tolerability, pharmacokinetics, pharmacodynamics, and clinical activity of L218CAR19 in patients with CD19-positive B-cell lymphomas who have received at least two lines of systemic treatment[1][3]. The primary completion date for this trial is expected to be December 31, 2025[3]. The study is being conducted by Henan Cancer Hospital, which is both the originator and active organization behind this therapy[2][3]. ## Target Population L218CAR19 is specifically designed for patients with: - CD19-positive B-cell lymphoma - Relapsed or refractory disease - Prior treatment with at least two lines of systemic therapy[1][3] This therapy represents part of the growing field of CAR-T cell immunotherapies that have shown promising results in treating various hematological malignancies, particularly those involving B cells that express the CD19 antigen.
