L523S is an immunogenic lung cancer antigen and oncofetal protein that has been investigated as a therapeutic cancer vaccine. It is delivered either as a recombinant DNA plasmid (pVAX/L523S) or via a replication-defective adenoviral vector (Ad/L523S), both encoding the human L523S protein. Upon administration, these vaccines express the L523S antigen in vivo, which may stimulate antibody and cytotoxic T lymphocyte responses against tumor cells expressing L523S. The primary indication explored for this therapy has been non-small cell lung cancer. Development was initially led by GSK and Corixa Corp., with clinical trials evaluating safety and immunogenicity in early-stage non-small cell lung cancer patients[1][2][3][5].
