LA-057-MMAE is a first-in-class antibody-drug conjugate (ADC) targeting the leukocyte-associated immunoglobulin-like receptor 1 (LAIR1). LAIR1 is an immune suppressive receptor expressed on immune cells and acute myeloid leukemia (AML) blasts, but minimally on hematopoietic stem cells, making it an attractive target for AML therapy. LA-057-MMAE consists of an anti-LAIR1 antibody conjugated to monomethyl auristatin E (MMAE), a potent cytotoxic agent, via a cleavable linker. The ADC binds strongly to human LAIR1, is efficiently internalized, and delivers the MMAE payload to induce cytotoxicity in AML cells. Preclinical studies have shown high affinity (3.9 nM) binding to LAIR1, efficient internalization (about 70% in 4 hours), and potent in vitro cytotoxicity with sub-nanomolar IC50 values against AML cell lines. In animal models, LA-057-MMAE caused complete tumor regression in 100% of AML xenograft mice at certain doses, indicating strong antitumor efficacy. The drug is distinct from existing LAIR1 monoclonal therapies and is the first ADC targeting LAIR1 specifically developed for AML[1][3].
