Overview

LA3IK is a first-in-class, MIEN1-based hexapeptide (chemically identified as Ac-LAIAVK-NH2) developed by the University of North Texas Health Science Center. Derived from the conserved regions of the Migration and Invasion Enhancer 1 (MIEN1) protein, LA3IK acts as a therapeutic peptide designed to inhibit cancer cell migration, invasion, and epithelial-to-mesenchymal transition (EMT). Mechanistically, LA3IK binds to MIEN1 and disrupts Epidermal Growth Factor (EGF)-driven oncogenic signaling. Specifically, it selectively interferes with the heterodimerization of Epidermal Growth Factor Receptor (EGFR) and ERBB2 (HER2), thereby reducing EGFR phosphorylation and suppressing downstream signaling pathways involving NF-κB, Src, and STAT3. LA3IK has demonstrated preclinical efficacy in breast, prostate, and colorectal cancer models, with its D-isomer (D-LA3IK) showing enhanced stability and pharmacokinetic properties.
