Labasiran (DV3396) is a GalNAc-conjugated small interfering RNA (siRNA) developed by Novo Nordisk, originally discovered by Dicerna Pharmaceuticals using its proprietary GalXC platform. It is designed to treat non-alcoholic steatohepatitis (NASH), also known as metabolic dysfunction-associated steatohepatitis (MASH), by targeting the mRNA of the HSD17B13 (Hydroxysteroid 17-Beta Dehydrogenase 13) gene. Genetic evidence suggests that individuals with loss-of-function mutations in HSD17B13 are protected against the progression of liver diseases, including NASH and cirrhosis. Labasiran mimics this protective effect by silencing the gene in hepatocytes, thereby reducing liver inflammation and preventing the progression of fibrosis. The drug is administered subcutaneously and is currently being evaluated in Phase 2 clinical trials.
