labetuzumab govitecan

**Labetuzumab govitecan** is an investigational, humanized anti-CEACAM5 antibody-drug conjugate developed by Immunomedics. It couples the anti-carcinoembryonic antigen-related cell adhesion molecule 5 antibody labetuzumab to the topoisomerase I inhibitor payload SN-38, the active metabolite of irinotecan, through a proprietary cleavable linker. The antibody targets CEACAM5-expressing tumor cells, particularly colorectal cancer, to concentrate and release SN-38 in the tumor microenvironment, resulting in topoisomerase I inhibition and cytotoxic DNA damage. It was evaluated in heavily pretreated metastatic colorectal cancer in Phase I/II studies, but the clinical development program was not advanced to registration and is considered discontinued.
