Overview

This combination therapy is a multi-component regimen being evaluated for the first-line treatment of *Helicobacter pylori* infection. It consists of lacidophilin (a probiotic containing *Lactobacillus acidophilus*), vonoprazan (a potassium-competitive acid blocker), and amoxicillin (a beta-lactam antibiotic). Vonoprazan suppresses gastric acid production by inhibiting the gastric H+/K+-ATPase pump more potently and for longer durations than traditional proton pump inhibitors, creating an optimal environment for amoxicillin's bactericidal activity. Amoxicillin acts by inhibiting bacterial cell wall synthesis through binding to penicillin-binding proteins. Lacidophilin is added to potentially enhance eradication rates and mitigate antibiotic-associated gastrointestinal side effects by modulating the gut microbiota. The regimen is currently being studied in Phase 4 clinical trials led by The First Affiliated Hospital of Nanchang University in China.
