Overview

Lacticaseibacillus rhamnosus DY801 is an investigational live biotherapeutic product, specifically a novel strain of Lactobacillus isolated from the fecal samples of rectal cancer patients who did not experience acute chemoradiotherapy-induced intestinal injury (ACRIII). This strain is being studied for its potential to mitigate ACRIII in patients undergoing chemoradiotherapy for colorectal cancer. The proposed mechanism of action involves the synthesis of methionine by the bacterium's metB gene. This microbial-derived methionine modulates the methionine metabolism in the host's gut lymphoid tissue inducer (Lti) cells. This leads to increased intracellular S-adenosylmethionine and subsequent histone H3 lysine 4 trimethylation (H3K4me3), an epigenetic modification. These changes result in the suppression of pro-inflammatory cytokines, specifically interleukin-17A (IL-17A) and interleukin-22 (IL-22), ultimately reducing the severity of intestinal injury without compromising the effectiveness of the cancer therapy.
