Overview

Lacticaseibacillus rhamnosus L34 (formerly Lactobacillus rhamnosus L34) is a probiotic strain isolated from infant feces. It is a vancomycin-resistant, Gram-positive, facultative anaerobic bacterium. The L34 strain has demonstrated ability to modulate inflammatory responses in the gut epithelium, particularly by suppressing Clostridium difficile-induced IL-8 and GM-CSF production in colonic cells through mechanisms involving inhibition of NF-κB activation. The active immunomodulatory factors produced by L34 are heat-stable and high molecular weight (>100 kDa), but their exact molecular identity is not specified. L34 does not directly inhibit C. difficile growth or toxin production, but instead exerts anti-inflammatory effects, suggesting potential for prevention or adjunct treatment of C. difficile-associated disease (CDAD). The strain is considered a candidate for probiotic development due to its safety profile and functional effects in vitro, but clinical evidence in humans is limited[1].
