Lactobacillus acidophilus CICC 6074 S-layer protein is a protein therapeutic under investigation for its potential anticarcinogenic activity, specifically against human colon cancer HT-29 cells. It has been shown to significantly suppress the proliferation of these cancer cells by arresting the cell cycle in the G1 phase. This cell cycle arrest is mediated through the upregulation of tumor suppressor proteins p53, p21, and p16, and the downregulation of cell cycle regulators CDK1 and cyclin B. Furthermore, this S-layer protein induces apoptosis in HT-29 cells via both the death receptor apoptotic pathway and the mitochondrial pathway. It also impedes cell invasion by inhibiting the PI3K/AKT signaling pathway and FasL. These findings suggest its potential as an anticarcinogenic agent.
