Overview

CKDB001 (*Lactobacillus delbrueckii* subsp. *lactis* CKDB001) is a probiotic strain developed for the treatment of cognitive impairment and neurodegenerative diseases such as Alzheimer's disease. As a live biotherapeutic, it functions through the gut-brain axis to modulate the intestinal microbiome and increase the production of neuroprotective metabolites, including tryptophan-derived indoles and short-chain fatty acids. Its neuroprotective mechanisms include the downregulation of hippocampal glycogen synthase kinase-3 (GSK-3) signaling, reduction of tau phosphorylation, and restoration of acetylcholinesterase (AChE) activity. Additionally, CKDB001 enhances gut barrier integrity by upregulating tight-junction proteins, which may help mitigate the systemic inflammation associated with cognitive decline. In preclinical models, its efficacy in improving spatial working memory has been shown to be comparable to that of donepezil.
