Overview

Lactobacillus johnsonii extracellular vesicles (EVs) are nano-sized, membrane-bound vesicles derived from the probiotic bacterium Lactobacillus johnsonii, particularly the N6.2 strain. These EVs encapsulate bacterial proteins, nucleic acids, and metabolites, including key active proteins like GroL (a chaperonin), GAPDH, and the Sdp-SH3b2 domain. They function as immunomodulators by promoting M2 macrophage polarization, regulating the Th17/Treg balance, and enhancing gut barrier homeostasis. Recent research highlights their ability to mitigate epithelial cuprotosis in ulcerative colitis by interacting with the host copper importer SLC31A1 to reduce intracellular copper accumulation. They are currently being investigated in academic settings for potential therapeutic applications in inflammatory bowel disease, osteoarthritis, and viral infections.
