Overview

Lactobacillus paracasei KKA101 is a specific strain of lactic acid bacteria, derived from Lactobacillus paracasei BL23, that serves as a live biotherapeutic delivery vehicle and a probiotic candidate. Developed and studied by researchers at institutions such as the Karolinska Institutet and the University of Tartu, KKA101 has been utilized as a base strain for engineering the secretion and surface display of therapeutic molecules, including pentameric glucagon-like peptide-1 (GLP-1) analogues for the treatment of type 2 diabetes and single-domain antibodies (VHH) for neutralizing Clostridium difficile toxins. In preclinical studies using diabetic Goto-Kakizaki (GK) rats, the non-engineered KKA101 strain (often used as a negative control) demonstrated an inherent ability to significantly lower blood glucose levels, suggesting that the strain itself possesses intrinsic anti-diabetic properties. Its mechanism of action likely involves the modulation of the gut microbiota and the production of metabolic byproducts, such as short-chain fatty acids, that improve glucose homeostasis.
