Overview

*Lactobacillus paracasei* KKA394 is a specific strain of probiotic bacteria investigated as a live biotherapeutic delivery vehicle for therapeutic proteins. In research contexts, it has been genetically engineered to express and secrete a pentameric analogue of Glucagon-like peptide-1 (GLP-1) for the treatment of type 2 diabetes. The strain is designed to deliver the GLP-1 analogue in situ to the intestinal mucosa, where it is cleaved by endogenous trypsins into an active monomeric form. This active GLP-1 then stimulates insulin secretion from pancreatic cells in a glucose-dependent manner. While the strain itself has demonstrated intrinsic glucose-lowering properties in diabetic GK (Goto-Kakizaki) rat models, the engineered version aims to provide a sustained insulinotropic effect to normalize blood glucose levels.
