Lactobacillus paracasei KKA403 is a specific strain of lactic acid bacteria investigated as a live biotherapeutic product (LBP) and a delivery vehicle for therapeutic proteins. In preclinical research, it has been engineered to express and secrete a pentameric glucagon-like peptide-1 (5×GLP-1) analogue for the treatment of type 2 diabetes. The strain is designed to survive the gastrointestinal environment and deliver active GLP-1 monomers to the intestinal mucosa following trypsin-mediated cleavage of the secreted pentamer. Preclinical studies in Goto-Kakizaki (GK) rats have demonstrated that the KKA403 strain itself possesses intrinsic glucose-lowering properties independent of the engineered payload, suggesting its potential as a microbiome-based therapy for metabolic disorders.
