Overview

Lactobacillus plantarum IMB19 (Lp IMB19) is a human-associated live biotherapeutic product (LBP) developed by ImmunoBiome Inc. for the treatment of various solid tumors. Identified using the AVATIOME screening model, Lp IMB19 modulates both innate and adaptive immunity to enhance anti-tumor responses. Its mechanism of action is primarily driven by a capsular component, a rhamnose-rich hetero-polysaccharide (RHP), which acts as a TLR2 agonist. This agonism targets macrophages, polarizing them toward an inflammatory phenotype and subsequently increasing the activity of CD8+ IFNγ+ T-cells. Furthermore, Lp IMB19 and its RHP component induce iron sequestration by activated macrophages within the tumor microenvironment, thereby depriving tumor cells of essential iron and suppressing their proliferation. Preclinical studies have demonstrated its efficacy in models of melanoma, renal cancer, non-small cell lung cancer (NSCLC), and urinary bladder cancer, particularly when used in combination with immune checkpoint inhibitors such as anti-PD-L1.
