Lactobacillus rhamnosus GG + peanut oral immunotherapy, also known as PPOIT (probiotic and peanut oral immunotherapy), is a combination immunotherapy developed by the Murdoch Children's Research Institute (MCRI) for the treatment of peanut allergy in children. [6, 8] The therapy consists of the daily oral administration of the probiotic *Lactobacillus rhamnosus* GG (strain CGMCC 1.3724/ATCC 53103) combined with peanut oral immunotherapy (OIT). [10, 13, 17] The treatment is designed to modulate the immune system's allergic response to peanuts, aiming to induce long-term, sustained unresponsiveness (remission) rather than just temporary desensitization. [2, 5, 7] This immunomodulatory effect involves reducing peanut-specific IgE levels and rewiring T-helper 2 (Th2) gene networks associated with the allergic response. [2, 4] The technology has been licensed to Prota Therapeutics, which is developing a high-dose version as PRT120 and preparing for Phase 3 trials. [3, 9, 20]
