Overview

*Lactococcus lactis* expressing Stat-CPSC5 is a bioengineered live biotherapeutic product (probiotic) designed for the precision targeting and elimination of *Fusobacterium nucleatum*, a bacterial pathogen strongly associated with the development and progression of colorectal cancer (CRC). The therapy utilizes a strain of *Lactococcus lactis* (typically MG1363) engineered to secrete a guided antimicrobial peptide (gAMP) known as Stat-CPSC5. This gAMP construct consists of a Statherin-derived guide peptide (Stat) that specifically binds to the FomA porin on the outer membrane of *F. nucleatum*, linked to a synthetic antimicrobial peptide (CPSC5). By delivering the antimicrobial payload directly to the surface of the target pathogen, the probiotic achieves selective depletion of *F. nucleatum* while preserving the diversity and richness of the surrounding gut microbiota. This targeted approach aims to mitigate the pro-tumorigenic and pro-inflammatory environment promoted by *F. nucleatum* in the colon, potentially serving as a therapeutic or preventive strategy for CRC.
