LAD328 (formerly known as SAR441121) is a trivalent nanobody being developed by Almirall S.A. under a licensing agreement with Sanofi. It is designed to simultaneously inhibit interleukin-17A (IL-17A) and interleukin-17F (IL-17F), two key pro-inflammatory cytokines that are significantly upregulated in various chronic inflammatory skin diseases. By targeting both isoforms, LAD328 aims to provide enhanced therapeutic efficacy compared to selective IL-17A inhibitors. The molecule is engineered as a trivalent single-domain antibody, which includes two domains for cytokine neutralization and a third domain that binds to human serum albumin to extend its circulating half-life. LAD328 is currently in Phase 2 clinical development for the treatment of moderate-to-severe hidradenitis suppurativa (HS), a debilitating inflammatory condition characterized by painful nodules and abscesses.
