Ladiratuzumab vedotin is an investigational antibody-drug conjugate (ADC) composed of a humanized monoclonal antibody targeting the zinc transporter protein LIV-1 (solute carrier family 39 member 6, SLC39A6), which is highly expressed in several solid tumors including breast cancer. The antibody is conjugated to a cytotoxic payload—monomethyl auristatin E (MMAE)—via a cleavable linker. Upon binding to LIV-1 on tumor cells, the ADC is internalized and releases MMAE inside the cell, leading to microtubule disruption, cell cycle arrest at G2/M phase, and apoptosis. Ladiratuzumab vedotin has shown activity as monotherapy and in combination with pembrolizumab in early-phase clinical trials for metastatic triple-negative breast cancer (TNBC) and other solid tumors expressing LIV-1[1][3][4][6][9].
