Ladiratuzumab vedotin + pembrolizumab is a combination therapy consisting of an antibody-drug conjugate (ladiratuzumab vedotin) and an immune checkpoint inhibitor (pembrolizumab). Ladiratuzumab vedotin targets LIV-1, a protein highly expressed in breast cancer cells, delivering the cytotoxic agent monomethyl auristatin E (MMAE) which disrupts microtubules and induces cell cycle arrest and apoptosis. Pembrolizumab is a humanized monoclonal antibody that blocks PD-1, enhancing T-cell responses against tumor cells. The combination leverages complementary mechanisms - LV's direct cytotoxic effects and induction of immunogenic cell death, which creates a favorable microenvironment for enhanced anti-PD-L1 activity from pembrolizumab.
