Ladostigil is a novel neuroprotective small molecule under investigation for the treatment of neurodegenerative disorders such as Alzheimer's disease and mild cognitive impairment. It was developed by structural modification of rasagiline and combines the mechanisms of action of both cholinesterase inhibitors (like rivastigmine) and monoamine oxidase B inhibitors (like rasagiline). Ladostigil acts as a reversible inhibitor of acetylcholinesterase and butyrylcholinesterase and an irreversible inhibitor of monoamine oxidase B. In addition to its neuroprotective effects, it enhances expression of neurotrophic factors such as GDNF and BDNF and may promote neurogenesis. Preclinical studies suggest antidepressant-like effects that could be beneficial in treating comorbid depression or anxiety in patients with neurodegenerative diseases[3][8][9].
