LADR-9 (PP072) is a preclinical investigational oncology drug candidate developed by LadRx Corporation and its subsidiary Centurion BioPharma. It is an albumin-binding prodrug that utilizes the proprietary Linker Activated Drug Release (LADR) technology platform. The drug consists of an ultra-high potency maytansinoid (maytansine) payload conjugated to a linker designed to bind to circulating albumin in the bloodstream. This small molecule drug conjugate is engineered to release the cytotoxic payload selectively at tumor sites in a pH-dependent manner. As a microtubule inhibitor, the released maytansine targets tubulin to induce cell death. Preclinical studies have demonstrated antitumor efficacy in various solid tumor models, including lung, breast, ovarian, renal cell, and head and neck cancers.
