Overview

LAE113 is a novel humanized IgG4 bispecific antibody (bsAb) targeting the immune checkpoints TIGIT (T-cell immunoreceptor with Ig and ITIM domains) and PVRIG (Poliovirus receptor-related immunoglobulin domain-containing protein). Developed by Laekna Therapeutics, LAE113 is designed to enhance anti-tumor immunity by simultaneously blocking two distinct inhibitory pathways within the PVR/PVRL family that suppress T cell and NK cell functions. The molecule is constructed using a humanized sequence from a lead anti-TIGIT monoclonal antibody and an anti-PVRIG single-domain antibody (sdAb). Preclinical data indicate that LAE113 is a potent signaling blocker that outperforms single-target competitors and demonstrates synergistic effects when combined with anti-PD-L1 therapies, such as LAE005.
