LAE119 is a novel, potent, and selective small molecule inhibitor of poly(ADP-ribose) polymerase 1 (PARP1), developed by Laekna. It acts as both a PARP1 enzymatic inhibitor and a strong PARP1-DNA trapper, demonstrating over 1000-fold selectivity for PARP1 DNA trapping activity compared to PARP2. Preclinical studies show that LAE119 has an extremely long residence time on the PARP1 enzyme in both biochemical and cellular assays. It exhibits robust anti-tumor efficacy in BRCA2-deficient xenograft models (such as DLD-1 and MDA-MB-436) with minimal hematologic toxicity. Notably, it retains activity even in tumor cells with low levels of PARP1 protein. The mechanism of action is based on synthetic lethality targeting DNA repair pathways in cancer cells[1][3][5].
