LAE120 is a novel, highly potent, allosteric inhibitor of ubiquitin-specific protease 1 (USP1), developed by Laekna for the treatment of advanced solid tumors. It features a unique chemical structure distinct from other USP1 inhibitors and demonstrates robust tumor inhibitory activity in preclinical xenograft models, including those with BRCA mutations and Bcr-Abl expression. LAE120 has shown efficacy both as a single agent and in combination with PARP inhibitors such as olaparib, particularly in homologous recombination deficiency (HRD) cancers. Preclinical studies indicate favorable pharmacokinetics, good therapeutic windows, and tolerability without significant toxicities. The drug is currently advancing through early clinical development following FDA approval of its Investigational New Drug (IND) application[1][2][3][4][5].
