LAE123 is an internally discovered dual antagonistic monoclonal antibody developed by Laekna. It potently blocks both activin receptor type IIA (ActRIIA) and activin receptor type IIB (ActRIIB), inhibiting their interaction with ligands such as myostatin, various activins (Activin A, B, AB, C, E), and growth/differentiation factors (GDF3 and GDF11). Dysregulation of ActRII signaling is implicated in diseases including muscular atrophy, obesity, sarcopenia, pulmonary arterial hypertension (PAH), spinal muscular atrophy (SMA), and other severe conditions. Preclinical studies have shown that LAE123 has superior potency in cell-based assays and favorable pharmacokinetics and efficacy in animal models[2][3][4][5].
