Lafutidine is a second-generation histamine H2 receptor antagonist used to treat gastrointestinal disorders such as peptic ulcers and gastroesophageal reflux disease (GERD). It acts primarily by inhibiting gastric acid secretion through antagonism of the histamine H2 receptor. Additionally, lafutidine exhibits a multimodal mechanism of action: it activates calcitonin gene-related peptide, stimulates nitric oxide production and regulates gastric mucosal blood flow, increases somatostatin levels (further reducing acid secretion), promotes mucin generation in the stomach lining, inhibits neutrophil activation to prevent inflammation-induced injury, and blocks *Helicobacter pylori* attachment to gastric cells. Its gastroprotective effects are also attributed to promoting restitution of damaged mucosa via capsaicin-sensitive afferent nerves rather than direct epithelial protection[5][6][7].
