LAG3-LaIL2 is a precision immunotherapeutic fusion protein designed to selectively deliver a low-affinity interleukin-2 (IL2) variant to LAG3-expressing exhausted CD8+ tumor-infiltrating lymphocytes (TILs). By targeting the Lymphocyte-activation gene 3 (LAG3) receptor, which is highly enriched on exhausted T cells within the tumor microenvironment, the drug aims to overcome T cell exhaustion—a major barrier to effective antitumor immunity. Mechanistically, LAG3-LaIL2 restores IL2R-JAK3-STAT5 signaling by upregulating the high-affinity IL2 receptor subunit CD122 (IL-2Rβ), thereby rejuvenating TIL functionality and preventing terminal exhaustion. This targeted approach enhances therapeutic efficacy while potentially improving safety by restricting IL2 exposure to non-target cells, thereby avoiding the systemic toxicities often associated with wild-type IL2 therapy.
