Overview

LAG525 + carboplatin is an investigational combination therapy for cancer, specifically studied in advanced triple-negative breast cancer (TNBC). LAG525 (also known as ieramilimab or IMP701) is a monoclonal antibody targeting lymphocyte activation gene-3 (LAG-3), an immune checkpoint receptor on T-cells. By blocking LAG-3, LAG525 aims to enhance T-cell activation and anti-tumor immune responses. Carboplatin is a platinum-based chemotherapy agent that induces DNA crosslinking and apoptosis in rapidly dividing cells. The rationale for combining these agents is that chemotherapy like carboplatin may alter the tumor microenvironment to make it more susceptible to immunotherapy by increasing tumor immunogenicity[2][6][10]. This combination has been evaluated in phase II clinical trials but did not demonstrate significant efficacy in shrinking tumors or improving outcomes compared to other regimens[1][7][10].
